SRB LIQUID BROTH


USES


SRB liquid broth is a liquid medium in 10 ml sealed sterile vials, suitable for the detection, differentiation and estimation of Sulphate Reducing Bacteria.

In industrial systems, Sulphate Reducing Bacteria cause corrosion to the metallic surfaces where they are attached as sessile microbes.

The formation of insoluble sulphides, as by product of their metabolism, results in plugging and fouling of the systems they contaminate.

The detection and estimation of these bacteria is based in their ability to convert sulphate ion to sulphide which reacts with the ferrous ions to give a black color.

Sulphate and ferrous ions are present in the liquid medium, together with the basic ingredients necessary to support the bacterial growth.


 


 

RECIPE DESCRIPTION


The liquid medium is derived from the Postgate Medium F recipe, which in turn is the iron-sulphite agar, widely used in food microbiology, supplemented with lactate and magnesium to support the growth of desulfovibrios and some other desolfotumacula genera (Mara & Williams, 1970).



DETECTION OF SRB


The simplest way to use liquid media to estimate the concentration of Sulphate Reducing Bacteria in a water sample is the extinction-dilution technique.

A more precise approach is based on the MPN (Most Probable Number) techniques.

A detailed procedure for both methods is available on request.


 

STORAGE


Store in a cool place at not more than 25 °C to not reduce the shelf life.



 

SAFETY PRECAUTIONS AND DISPOSAL


SRB liquid medium is for laboratory use only.

Do not inject in humans.

Blackened and used vials can be disposed off after autoclave sterilization or chemical sterilization (e.g. inject 1 ml of commercial bleach liquor into each blackened or used vial).

PHENOL RED LIQUID BROTH

 

USES


Phenol red liquid broth is a 9 ml liquid medium in sealed sterile vials, suitable for the detection, differentiation and estimation of acid producing aerobic bacteria and/or total aerobic bacteria.

In industrial systems, acid producing aerobic bacteria cause corrosion and/or fouling over the metallic surfaces where they are attached as sessile microbes.

The detection and estimation of these bacteria is based in their ability to cause a change in colour, from red to yellow, of the liquid medium.

Phenol red dye, as pH indicator, is present in the liquid medium together with the basic ingredients necessary to support the bacterial growth.

 


RECIPE DESCRIPTION


The liquid medium is derived from the classical Phenol red carbohydrate media (DIFCO MANUAL).



DETECTION OF AEROBIC BACTERIA

 

Injected vials that turn yellow after incubation indicate the presence of aerobic acid producing bacteria in the sample of tested water.

Injected vials that become turbid without change in colour indicate the presence of general aerobic bacteria (not acid producing) in the sample of tested water.
